Weiser River Rail Trail is a ride with no markings--you just follow the
old railbed. (Well, I guess they did have a couple of marked spots this year--I didn't get there to ride it this year--but my understanding was that there were a couple of bridges that were out or not safe or something, so there were marked routes around them.)
As long as people know in advance that there is a section that is not
marked, and it is that obvious, it shouldn't present a problem. But mile markers sure are nice--WRRT has them, and it is nice to be able to count them off. As long as you have either human beings (as you described) at beginning and end, or a big sign saying "no more markings for 10 miles" when you start on it and really obvious markings when you get to the end of it, it shouldn't be a problem.
