Working up hill will allow a horse to work against more resistance- his own weight. I am currently able to do this as often as I want. Mountains surround us; it is hard to find a trail that does not go up hill. I can't site a paper, or even quote exercise efficiency changes on even one horse, but my horses seem to be benefiting from the hill work. If one wanted to do real weight training with horses you could train them to pull a stone boat. The weight in the stone boat could then be adjusted so the horse did say 10 repetitions of 16.5 feet for the workout. The draft horses I watched in pulling contests surely seemed to have enough strength to satisfy anyone.
