Makes a huge difference. Only you know how much you can tolerate. I did my first 100 only a couple of weeks after tweaking my left knee extreme bowling... it swelled up and pained me big time. I wore a knee brace during the ride which helped and took lots of ibuprofen (and the occasional Vicodin) and made it through and had a great time. But I also am too stubborn at times and have a high tolerance for pain. What does Angie say? If you're gonna be stupid, you gotta be tough. I'm actually relaxing that philosophy nowadays. :) I want to be riding when I am REALLY old and grey! But you know what you can handle (or not). The MSM nowadays makes all the difference for me but some people can't tolerate it. I take 1000-2000 mg a day. Good luck whichever you decide to do, the 50 or 100.
