I think so, and it would certainly be my first priority if I had a "semi-pet" skunk going in and out of my house.  In south Texas when there was a really bad rabies outbreak many years ago, there was a program where an oral type of vaccination was used, they left food out all over south and southwest Texas for the coyotes and fox to eat, I would assume skunks would eat it also. 
 
Worth a try to find out from your local veterinarian to see if there's an oral "vaccine" that you could put out for the skunk.

When It was time, we released her here at the house as she is a great mouser and skunks usually only spray when they are threatened or frightened. "Daisy" has stuck around the house and is "fully-armed". She freely comes and goes through the cat door & has never sprayed. As you can see from these pictures, she has obviously adopted our dog as her "lover". We DO pet her and she has never bitten, but are still very wary just in case she were to pick up the rabies virus and become a carrier.
